Automated detection of protein unfolding events in atomic force microscopy force curves

Atomic force microscopy is not only a high-resolution imaging device but also a mechanical machine, which can be used either to indent or stretch (soft) biomaterials. Due to the statistical nature of such materials (i.e., hydrogels or polymers) hundreds of force-distance curves are required to describe their mechanical properties. In this manuscript, we present an automated system for polymer unfolding detection based on continuous wavelet analysis. We have tested the automated program on elastin, which is an important protein that provides elasticity to tissues and organs. Linear Dimensions of Adsorbed Semiflexible Polymers: What can be learned about their persistence length?

Conformations of partially or fully adsorbed semiflexible polymer chains are studied varying both contour length $L$, chain stiffness, $\ensuremath{\kappa}$, and the strength of the adsorption potential over a wide range. Molecular dynamics simulations show that partially adsorbed chains (with ``tails,'' surface attached ``trains,'' and ``loops'') are not described by the Kratky-Porod wormlike chain model. The crossover of the persistence length from its three-dimensional value (${\ensuremath{\ell}}_{p}$) to the enhanced value in two dimensions ($2{\ensuremath{\ell}}_{p}$) is analyzed, and excluded volume effects are identified for $L\ensuremath{\gg}{\ensuremath{\ell}}_{p}$. A Monte Carlo Study of Living Polymers in 2D: Effect of Small Chains on Static Properties

A slithering snake algorithm is combined with a binding and breaking chain algorithm to simulate the static behavior of living polymers according to Cates' description. It is shown that this simple two-dimensional simulation on a square lattice gives good agreement with the mean field theory. However, the large amount of small contour length chains for small values of the mean average length 〈L 〉 appears to be one of the reasons for the discrepancies observed between the simulated results and the mean field theory. This finding could explain disagreements between experimental observation and theory. Also, the results are not in favor of a swelling of the greater chains by the smaller one.
